The Arp2/3 activator WASH regulates a5b1-integrin-mediated invasive migration

چکیده
The actin cytoskeleton provides scaffolding and physical force to effect fundamental processes such as motility, cytokinesis and vesicle trafficking. The Arp2/3 complex nucleates actin structures and contributes to endocytic vesicle invagination and trafficking away from the plasma membrane. Internalisation and directed recycling of integrins are major driving forces for invasive cell motility and potentially for cancer metastasis. Here, we describe a direct requirement for WASH and Arp2/3-mediated actin polymerisation on the endosomal membrane system for a5b1 integrin recycling. WASH regulates the trafficking of endosomal a5b1 integrin to the plasma membrane and is fundamental for integrin-driven cell morphology changes and integrin-mediated cancer cell invasion. Thus, we implicate WASH and Arp2/3-driven actin nucleation in receptor recycling leading to invasive motility.

متن کاملIntegrin-mediated Adhesion Regulates Cell Polarity and Membrane Protrusion through the Rho Family of GTPases□V
Integrin-mediated adhesion is a critical regulator of cell migration. Here we demonstrate that integrin-mediated adhesion to high fibronectin concentrations induces a stop signal for cell migration by inhibiting cell polarization and protrusion. On fibronectin, the stop signal is generated through a5b1 integrin-mediated signaling to the Rho family of GTPases. Specifically, Cdc42 and Rac1 activa...
متن کاملLoss of Scar/WAVE Complex Promotes N-WASP- and FAK-Dependent Invasion
BACKGROUND The Scar/WAVE regulatory complex (WRC) drives lamellipodia assembly via the Arp2/3 complex, whereas the Arp2/3 activator N-WASP is not essential for 2D migration but is increasingly implicated in 3D invasion. It is becoming ever more apparent that 2D and 3D migration utilize the actin cytoskeletal machinery differently. RESULTS We discovered that WRC and N-WASP play opposing roles ...

Both the transforming growth factor b (TGF-b) and integrin signalling pathways have well-established roles in angiogenesis. However, how these pathways integrate to regulate angiogenesis is unknown. Here, we show that the extracellular matrix component, fibronectin, and its cellular receptor, a5b1 integrin, specifically increase TGF-b1and BMP-9-induced Smad1/5/8 phosphorylation via the TGF-b su...
متن کاملDrosophila WASH is required for integrin-mediated cell adhesion, cell motility and lysosomal neutralization.
The Wiskott-Aldrich syndrome protein and SCAR homolog (WASH; also known as Washout in flies) is a conserved actin-nucleation-promoting factor controlling Arp2/3 complex activity in endosomal sorting and recycling. Previous studies have identified WASH as an essential regulator in Drosophila development. Here, we show that homozygous wash mutant flies are viable and fertile. We demonstrate that ...
